(23) LPS - PANCREATIC ENZYMES Flashcards
LPS Systematic name:
Triacylglycerol Acylhydrolase
- Earliest / most specific pancreatic marker
- Hydrolyzes the ester linkages of fats to produce alcohols & fatty acids
LPS (Lipopolysaccharide)
main source of LPS
Pancreas
o TAGs as substrate (hydrophobic – insoluble to H2O)
o 2-monoglyceride & Fatty acids as product (we measure the FA liberated from TAGs)
↑LPS activity = ↑fatty acids liberated from TAGs
breaks down fats (TAG to monoglyceride)
Lipase
Acute pancreatitis:
LPS rises: ____ after onset
Peaks: ____ after onset
Remains elevated for: _____
Acute pancreatitis:
LPS rises: 6hrs after onset
Peaks: 24hrs after onset
Remains elevated for: 7 days / 1 wk
Reference method for LPS
Cherry Crandall
Cherry Crandall Indicator
Phenolphthalein
Tietz indicator
Thymolphthalein + Veronal
Substrate of both Cherry Crandall & Tietz
50% Olive oil (triolein) – half is water bcos LPS is a hydrolase
Titrating agent of both Cherry Crandall & Tietz
0.4N NaOH
Cherry Crandall end color
Pink
Tietz end color
Blue
o Estimation of liberated fatty acids
o ↑TAGs = turbid soln
o Activity of LPS to triacyglycerol as substrate is inversely proportional to absorbance reading of soln.
▪ More active = ↑LPS activity to substrate = less turbid suspension = ↓absorbance
