unit 10 Flashcards
examples of slow virus diseases
SSPE –> measles (mutated m gene)
PRP –> rubella (congenital rubella)
Cancer –> HTLV-1
what is a slow virus disease
virus that has a prolonged incubation period in which it is slowly replicating without being detected
–> it will then gradually damage cells
prions
misfolded proteins that transmit their infectiousness to others
PrPc
normal protein isoform
PrPsc
infectious isoform
theory explaining the mode of replication of prions
infectious form binds to normal form and converts it through binding
how are prions typically transmitted
either acquired (GI tract/broken skin or contaminated surgical instruments)
inherited (passed down)
sporadic (mutations)
pathogenesis of the transmissible spongiform encephalopathies
PrPsc moves form PNS to CNS
and when it increases it will lead to amyloid formation and cause holes where neurons die
match the TSE with its natural host
scrapie –> goat/sheep
BSE –> cattle
CWD –> deer
Kuru –> humans
CJD/vCJD –> humans and cattle
symptoms and time of course of CJD and variant vCJD
sensory disturbances, memory loss, progressive dementia, spasms
–> main difference is that vCJD has earlier onset age and longer course
origin of vCJD
BSE (eating infected cattle)
laboratory tests used to diagnose CJD
serology (14-3-3 protein presence)
perfrom RT-Quic which test ability of sample to convert recombinant protein
