Write method for dehydrogenase enzyme practical

A
  1. Cut leaves
  2. Use pestle and mortar to bring leaves with some chilled isolation solution
  3. Filter liquid into beaker using funnel lined with muslin cloth
  4. Transfer to centrifuge tubes
  5. Centrifuge at high speeds for 10 mins
  6. Chloroplasts will gather at bottom as pellet
  7. Remove supernatant
  8. Resuspend pellet in fresh chilled isolation solution
  9. Store in ice bath
  10. Set up calorimètre with red filter
  11. 0 calorimètre with distilled water
  12. Set up test tube rack with a certain distance away from the lamp
  13. Switch lamp on
  14. Add 2cm3 of DCPIP and 2cm3 of chloroplasts extract
  15. Mix
  16. immediately take out sample and put in calorimètre
  17. Repeat every 2 mins for 10 mins at different distances away from the lamp
