Therapeutic Indix
Compares ED50 and LD50 (lethal dose). Large therapeutic TI = low risk for toxicity while small TI = risk for toxicity and need plasma monitoring.

Dose-response relationships
Can’t just increase dose and response…
Allergies are different; an exaggerated immune response.
Potency
How strong a drug is based on dose. Higher dose = lower potency, lower dose= higher potency.
Efficacy
Intensity of drug response… morphine works more intensely at reducing pain than ibuprofin so it has higher efficacy.
Down regulation
Concept of drug tolerance… need an increase in drug dose to get the same response.
Upregulation
Ex. Beta Blocker to decreases HR….up-regulation due to receptors requires pt to wean off meds.
Agonist
Binds to receptor appropriately
Partial agonist
Partially binds bud not fully so elicits a partial response.
Antagonist
Blocks what body would normally do
Agonist plus antagonist
Also has partial response like partial.
