Culture:
Blood Agar - alpha hemolytic colonies (draughtman’s colonies)-due to autolysin prod which lyses its own cell wall
Chocolate agar
Specific tests:
Oxidase –ve, Catalase –ve
Inulin fermentation +ve
Bile solubility +ve
Optochin sensitive
Pathogenic to mice (lethal)
case
A 60-year-old man
-high-grade fever with chills and rigors for the previous two days.
- mild chest pain and productive cough.
-X-ray chest showed consolidation in the right lower lobe.
- Gram staining showed pus cells with Gram positive, lanceolate- shaped diplococci.
AST:sensitive to penicillin
serology
counter-current immunoelectrophoresis,latex agglutination to demonstrate capsular antigens
