What are the Specimen to be collected:
Blood, Bronchoalveolar lavage
High risk group for CMV infections:
Organ transplants, immunodeficient persons and in premature infants
DIAGNOSTIC TESTS:
- Serological tests for confirmation,
- Isolation of viruses
Tissue culture and Immunofluorescence staining- human fibroblast cultures
Buffy Coat blood culture method using white blood cells
Cytomegalovirus-infected cells show characteristic abundant cytoplasm with central granular eosinophilic zone and eccentric, slightly concave nucleus (Wright-Giemsa staining)
inclusion bodies in the infected tissue-
owl’s eye intranuclear inclusion bodies
Serological tests:
CF- Complement fixation- Presumptive diagnosis
* IHA- Hemagglutination inhibition
* IF-Immunofluorescenceassayfordetectionofviral
antigens
* IgM, IgG antibodies against CMV– ELISA (very specific)
