Why is aseptic technique important in culturing bacteria?
-Keep apparatus and equipment free from micro organisms
-Prevent cost contamination of other bacteria to get into petri dish
-prevent cultured bacteria to get into environment
How to use aseptic before experiment
Area - wipe surface, wear glove, wash hands with soap or alcohol, Bunsen on full blast for ‘clean’ air
Media (petri dish) - autoclave
Reusable equipment - inoculating loop (things that scoop up bacteria from petri or environment) heated with Bunsen until red and cooled, glass wipe or autoclave
Single use plastic with equipment inside - radiation
How to do aseptic during experiment
Media (petri) - lift open lid just enough to wipe it with bacteria (prevent air born bacteria)
Inoculating loop - immediately go over Bunsen (kill bacteria after scooping)
How to do aseptic after culturing
-wipe down surfaces
-Remnant (used Petri dish) - seal lid with tape, prevent anaerobes since allow oxygen in (most anaerobes are human pathogens)
+under 25 degree (prevent human pathogens)
Remnant (petri used after cultured) - auto clave and bin
Autoclave
-A seal container that allow glass or metal equipment to be heated at 121 degree and under pressure to destroy any bacteria (including those resistant to boiling)
-disposable equipment (petri dish) sealed inside autoclavable plastic (high temp resist) and be binned
