Lincosamides Flashcards
What is the MOA of Clindamycin?
Binds to 50s ribosomal subunit and inhibits protein synthesis
What drug classes is Clindamycin similar to based on MOA:
Macrolides & Oxazolidiones (Linezolid)
Is Clindamycin bacteriostatic?
Yes
Why is Clindamycin use limited?
High risk of GI disturbances d/t C.Diff overgrowth
Blackbox warning for Clindamycin?
C.Diff overgrowth can cause Pseudomembranous colitis
PO absorption of Clindamycin:
Complete absorption. Highly protein bound
How is clindamycin distributed?
Distributes to pleural/peritoneal fluids with high concentrations in bile
Clindamycin & Pregnancy
Safe to use. Crosses placenta & excreted in breast milk
How is Clinda metabolized & excreted? Dose modifications required?
Metabolized in liver
Excreted in bile & urine
No dose adjustment unless severe impairments
Important Drug Interactions for Clinda:
- Erythromycin: (competes with Macrolides d/t mechanism of action)
- Kaolin Pectin
- Neuromuscular blockers
What is clindamycin used for?
Bacterial endocarditis
Septic shock (decreases toxin production)
PJP PNA
Drug resistant pneumococcal infections
