LETHAL EFFECTS OF TEMPERATURE Flashcards
What are the 2 methods used to compare organisms to elevated temperatures?
Thermal death point (TDP) and Thermal death time (TDT)
What is the Thermal Death Time?
TDT is the time required to kill a suspencion of cells at a given temperature
What is the Thermal Death Point?
TDP is the temperature at which an organism is killed in 10 minutes
What are the various factors that will greatly influence the temperature values
TDT/TDP
pH, moisture, composition of medium and age of cells
What are the 2 broth cultures used for the lethal effects of tenperature experiment?
Bacillus megatarium and Escherichia coli
How does the TDT differ from TDP?
The difference between time and point is that time is given temperature while point is in 10 mins
Why is bacillus megatarium not classified as a thermophile?
Because it has a strong endospore that can survive higher temps
Why do different microbes have different thermal death times?
Different bacterial species have different temp required for growth
What is the advantage of having a high thermal death time?
It kills bacteria faster
What is the importance of inoculating a control plate in this experiment?
The control plate with the bacteria not exposed to higher temperatures is used for comparison and to ensure that the bacteria is capable of bacteria
