Lecture 8 - Pyridoxal Phosphate Mechanism Flashcards
What is the first step of the PLP mechanism?
Amine group of amino acid attacks the aldehyde of PLP
What is the second step of the PLP mechanism?
Loss of water and rearrangement of double bonds
What is the third step of the PLP mechanism?
Addition of water across the C=N bond leaving an OH on the C
What is the fourth step of the PLP mechanism?
OH on the C forms C=O and dissociates itself from the N, releasing pyruvate
What is the fifth step of the PLP mechanism?
NH2 on the PLP goes through the reverse reaction, attacking the ketone group of alpha-ketoglutarate
What is the sixth step of the PLP mechanism?
Water is released producing a C=N bond
What is the seventh step of the PLP mechanism?
Rearrangement of electrons, followed by addition of water across the double bond
What is the eighth step of the PLP mechanism?
Formation of an aldehyde group on PLP and release of glutamate
What is the cofactor of step 1?
H+ out
