Tositumomab is a murine IgG2a lambda monoclonal antibody directed against the CD20 antigen, which is found on the surface of normal and malignant B lymphocytes. Iodine I 131 Tositumomab is a radio-iodinated derivative of Tositumomab that has been covalently linked to Iodine-131. I131 BEXXAR Tositumomab Dose & Administration

A

There are four components of administration in two discrete steps.
The Dosimetric Step:
- Administer about 450 mg of Tositumomab intravenously in 50 mL 0.9% Sodium Chloride over 60 minutes. Administer Iodine I131 Tositumomab, which contains 5 mCi of Iodine-131 and 35 mg of Tositumomab, intravenously in 30 mL 0.9% Sodium Chloride over 20 min.
The Therapeutic Step:
- Administer about 450 mg of Tositumomab intravenously in 50 mL 0.9% Sodium Chloride over 60 minutes. For administer Iodine I131, you need to calculate the dose by determining total body residence time and activity hours.

A

The iodide is primarily distributed within the extra-cellular fluid of the body and it is trapped by the thyroid. Trapped iodide is oxidized to iodine and incorporates so rapidly that the iodide trap of the thyroid contains less than 1 percent of free iodide. This process results in further concentration of iodine in the thyroid gland to about 500 times that in the blood.
