Exercise 13 Flashcards
Vegetative cell
Actively growing and metabolizing yeast or bacteria
Spore
Survival mechanism that packages DNA into an extremely resistant dormant cell type
Thick spore coat
Resistant to dehydration, radiation, heat, and toxic chemicals
Spores could survive hours of boiling and high doses of radiation that would kill humans
True
Spore metabolic activity
Low
Sporulation/sporogenesis
Process of generating spores triggered by poor conditions, low carbon and nitrogen
Occurs when favorable conditions return
Germination which regenerates the vegetative cell
Endospore
Spore inside dying bacterial cell
Exospore
A spore that has been released from a bacterial cell that lysed
3 spore genres
Clostridium, bacillus, sporosarcina (only cocci)
2 genera that impact healthcare field
Clostridium and bacillus, not killed with typical disinfectant
Endospore procedure
Airdry and heat fix
Primary stain: malachite green
Mordant: steam/heat to soften spore and allow edge in + fixes dye
Decolorizer: distilled water
Counterstain: safranin
Endospore stain result
Endospore green, rest of cell pink
Endospore stain result W/ out enough mordant
Endospore colorless, vegetative part of cell pink
Result with not enough decolorizer
Endosperm colorless, vegetative part of cells pink
