Explain the TLC method
- TLC is performed on a sheet of glass, plastic or aluminium foil coated w/absorbent material. Known as the stationary phase. Absorbent material can be silica gel, aluminium oxide or cellulose
- Small spot of solvent containing the sample is added to the plate, 1 - 1.5cm from bottom edge
- An appropriate solvent is poured into the glass to a depth less than 1cm. The plate with sample is put into the chamber, so bottom touches the solvent. Chamber is closed with a lid & left for a few minutes, so the solvent can be drawn up the plate by capillary action (mobile phase)
- When the solvent mixture has been drawn up closely to the top, it is taken out from the chamber
- Sample plate is dried off and observed in visible or UV light
- Visible spots are measured & compared with the other spots. The different analytes will move along the TLC plate at different rates
