Chromosome structure and function Flashcards
What are the roles of proteins in chromosomes?
- Package and unfold DNA
- Control DNA replication
- Maintain chromosome integrity
- Govern proper chromosome segregation
- Regulate gene expression
Whats a telomere?
DNA sequences at the end of linear chromosomes
What is the centromere?
DNA sequences on which the kinetochore assembles and mediates chromosome segregation
What is the kinetochore?
A protein complex that binds microtubules in the the mitotic spindle
What are telomeres replicated by?
Special DNA polymerase- telomerase
What is a nucleosome?
A core protein with DNA wound around it
What is a chromosome?
A highly coiled fibre of chromatin
What are the subunits of the nucleosomes called?
Core histones
What is H1 and what does it do?
Linker histone
Straps DNA onto histone octomers and limits the movement of DNA
What are transposons?
Repeated DNA sequences
mobile, genetic elements that jump around the genome
3 examples of transposons
- DNA transposons
- Retroviral retrotransposons
- Non-retroviral polyA retrotransposons
What does transposase do?
‘Cut and paste’ transposons - moving them around the genome
How do retroviral transposons behave?
Replicate via RNA intermediates- uses own reverse transcriptase- produce new DNA copes that integrate at new genomic locations
How do non-retroviral transposons behave? 2 examples of such transposons
Replicate via RNA intermediates- uses own reverse transcriptase
Human L1 elements
Human Alu elements
Mouse B1 element
What are possible implications of some L1 insertions?
Can disrupt genes- cause disease e.g haemophilia
