What does forward genetics involve?
Using mutagenesis to create random mutations- then identify the gene that underlies the resulting phenotype
What does a complementation test do?
Tests whether the mutations in 2 strains are in different genes
What does a linkage analysis do?
Tests if 2 genes are linked- tendency of genes to be inherited together because of their location
What are SNPS?
Genetic variation between individuals
How do mutations affect gene function?
- Changes in regulatory sequences
- Changes in non-coding sequences
- Changes in the coding sequence
What are the types of mutation?
- Amorphic- complete loss of function
- Hypomorphic- decrease in wildtype function
- Antimorphic- mutant proteins interfere with wild type proteins
- Hypermorphic- overexpression of the T.factor/ over activity of gene product
