Cholesterol drugs from PBL 4 Flashcards
HM-CoA reductase inhibitors
prevent formation of mevalonate in cholesterol synthesis
statins
Inhibits lipolysis in adipose tissue (catabolism of apo AI?)
reduces production of VLDL in liver
Niacin (vit B3)
PPAR agonist
upregulates lipoprotein lipase, apo AI, AII
downregulates apo CIII (a lipolysis inhibitor)
Decreases VLDL
Increases HDL
fibrates
Gemfibrozil
Fenofibrate
Blocks sterol transporter NPC1L1 in intestinal brush border
decreased delivery of intestinal cholesterol to liver.
Ezetimibe
Binds to bile acids in the intestine and forms a nonabsorbable complex that is excreted in feces
results in increased conversion of cholesterol to bile acids in the liver
increased demand for cholesterol in liver cells, resulting in a compensatory increase in hepatic uptake (and thus systemic clearance) of circulating LDL-cholesterol.
Colestipol
bile acid sequestrant
