Chapter 5- Protein methods Flashcards
The proteome is the entire set of ______ expressed and modified by a cell under a particular set of ___________ conditions
Proteins,Biochemical
Proteins can be purified on the basis of ________ in their ___________ properties
Differences,Chemical
Proteins can be purified according to what 4 things?
Solubility
Size
Charge
Binding affinity
How does Ion exchange Chromatography work?
It allows for separation according to charge, so when the protein flows through the tube the protein with same charge as the beads already in the tube will pass through and the oppositely charged ones will bind to the beads.
How does the model of Affinity Chromatography work?
There is glucose binding protein that a attaches to glucose residue on beads but when more glucose is released into the tube, those same glucose binding proteins are released.
What is gel electrophoresis?
Proteins migrate in an electrical field since they are charged and this process is when migration happens in a gel
How does isoelectric focusing work?
- this allows for separation of proteins in a gel basis of their relative amounts of acidic and basic amino acids.
what can the estrogen receptor be purified by?
Immunoprecipitation
