What are the 5 Is
Inoculation - to grow, or culture, microorganisms, one that introduces a tiny sample into a container of medium
Incubation - it is placed in a temperature controlled chamber to encourage multiplication
Isolation - requires that a small number of cells is inoculated into a relatively large volume or over a large area of medium
Inspection - observe the microorganism through a microscope
Identification - classifying one of the microorganisms as different from another, figuring out what each one is
Semisolid media
Solid media
Liquid media
Semisolid - prepared with a lower agar concentration of 0.2 to 0.5% and has a soft custard like consistency
Solid - used to grow microorganisms in their full physical form, prepare bacterial pure cultures, or isolate bacteria to study the colonies characteristics
Liquid - do not contain any traces of solidifying agents, like an agar or gelatin, and large growths of bacterial colonies can be observed
How can you tell if a culture is pure
You can tell a culture is pure if it only contains one single type of bacteria
A mixed culture has two or more types of bacteria
How do you calculate the total magnification of a microscope
Multiple the power of the ocular lens by the power of the objective lens
