Bone Disorders Flashcards
Alendronate
Aminobisphosphonates
- Inhibits Osteoclast Activity
Disrupts Mevalonate pathway leading to disruption of osteoclast activity, leading to apoptosis of osteoclasts
Risedronate
Aminobisphosphonates
- Inhibits Osteoclast Activity
Disrupts Mevalonate pathway leading to disruption of osteoclast activity, leading to apoptosis of osteoclasts
Pamidronate
Aminobisphosphonates
- Inhibits Osteoclast Activity
Disrupts Mevalonate pathway leading to disruption of osteoclast activity, leading to apoptosis of osteoclasts
Zoledronate
Aminobisphosphonates
- Inhibits Osteoclast Activity
Disrupts Mevalonate pathway leading to disruption of osteoclast activity, leading to apoptosis of osteoclasts
Etidronate
Non-Aminobisphosphonates
- Inhibits Osteoclast Activity
Increases accumulation of cytotoxic metabolites within osteoclasts, leading to osteoclast cell death
Clodronate
Non-Aminobisphosphonates
- Inhibits Osteoclast Activity
Increases accumulation of cytotoxic metabolites within osteoclasts, leading to osteoclast cell death
Raloxifene
Selective Estrogen Receptor Modulators (SERM)
- Stimulates Osteoblasts
- Inhibits Osteoclasts
Teriparatide
PTH
- Increases Serum Calcium
- Decreased Serum Phosphate
- Indirect: Increased RANKL = Increase Osteoclast activity
Low Intermittent Dose:
- Increase Osteoblasts
- Inhibit apoptosis of Osteoblast
- Stimulate IGF (Promotes Bone Formation)
- Kidney Effects:
–> Increased Ca2+ and Mg2+ reabsorption
–> Decreased Phosphate, amino acids, bicarbonate, Na+, Cl-. and Sulfate reabsorption
–> Stimulation of Vitamin D production
Calcitriol
Vitamin D
- Increases Calcium absorption/reabsorption
- Stimulates Osteoblast/Osteoclast
- Decreases PTH
Ergocalciferol
Vitamin D
- Increases Calcium absorption/reabsorption
- Stimulates Osteoblast/Osteoclast
- Decreases PTH
Calcipotriene
Vitamin D
- Increases Calcium absorption/reabsorption
- Stimulates Osteoblast/Osteoclast
- Decreases PTH
Doxercalciferol
Vitamin D
- Increases Calcium absorption/reabsorption
- Stimulates Osteoblast/Osteoclast
- Decreases PTH
Paricalcitol
Vitamin D
- Increases Calcium absorption/reabsorption
- Stimulates Osteoblast/Osteoclast
- Decreases PTH
Denosumab
Inhbibits RANKL
- Prevents RANK (On osteoblasts) from binding to RANKL (On osteoclasts)
- Prevents differentiation, activation, and survival of Osteoclasts
Decreased Bone Resorption
Salcatonin
Calcitonin
- Decreases reabsorption of calcium and phosphate in the kidney
- Inhibits osteoclast activity
–> Inhibits bone resorption
