Antipsychotics Flashcards
chlorpromazine, fluphenazine
phenothiazines (typical D2 antagonists)
side effects: dry mouth (mAChR), postural hypotension (alpha2)
may inc negative symptoms, parkinsonism, cognitive deficit
haloperidol, pimozide
butyrophenones (typical D2 antagonists)
not as potent, better tolerated
atypical antipsychotics (5HT2 and D2 antagonists)
clozapine, risperidone, aripriprazole
side effects: weight gain, insomnia, sedation
aripriprazole agonises in mesocortical pathway (dec neg symptoms) and antagonises in mesolimbic pathway (dec pos symptoms)
5HT2C side effect
weight gain (stop satiety blockade)
alpha1AR side effect
postural hypotension
H1 side effect
sedation
mAChR side effect
dry mouth, dry eyes etc
5HT2A side effect
extrapyrimidal
atypical vs typical
atypical has less extrapyrimidal side effects (FAST OFF - some binding retained in nigrostriatal pathway)
atypical has more action on negative side effects
positive symptoms
inc mesolimbic DA
negative symptoms
dec mesocortical DA
schizophrenia pathology
hyperdopaminergic
hyperserotinergic
hypoglutametergic
neg side effects in tuberoinfundibular pathway
hyperprolactinaemia
