TCA MOA
tricyclic antidepressants
block reuptake of Noradrenaline
blocks 3 other receptors:
muscarinic
H1
alpha 1-adrenoreceptors
TCA side effects
blockade of muscarinic receptors
•dry mouth, blurred vision, urinary retention, constipation
– blockade of H1 receptors
•sedation
– blockade of alpha1‐adrenoceptors
•orthostatic hypotension
also get arrhythmias/heart problems (its cos it raises noradrenaline levles in brain but the heart as well, driving heart harder)
desipramine
tricyclic antidepressants
SSRI MOA
specifically block the re‐uptake of 5HT
SSRI side effect
GI upset
cos 90% of the bodys 5HT is in the enterochromaffin cells in the GIT
SSRI contraindication
st johns wort
fluoxetine
SSRI
Irreversible MAOa inhibitor MOA
MAOa breaks down NA and 5HT
so u inhibit it permanently
enzyme activity returns with resynthesis of new enzymes 2-3 wks
Irreversible MAOa inhibitor side effects
cheese reaction
insomnia
postural hypotension
atropine-like side effects
sexual dysfunction (e.g. low libido)
tranylcypromine

bind to MAOa reversibly
no cheese reaction
