A

so that when they ligate in they don’t cut the back-bone but they cut the GFP

Why would you see all green colonies after Golden Gate?

A

The Bsa1 didn’t cut because you forgot to add it or you did something to deactivate it (heating it)

A

Because it cut it, but the ligation didn’t work.

  • DNA strands are pulled through a nanopore
  • Miniscule changes in electric current are converted into base reads for each nucleotide
  • Produces longer reads then competing technologies
