A
so that when they ligate in they don’t cut the back-bone but they cut the GFP

Why would you see all green colonies after Golden Gate?
A
The Bsa1 didn’t cut because you forgot to add it or you did something to deactivate it (heating it)

Why would you see no colonies after Golden Gate?
A
Because it cut it, but the ligation didn’t work.

What are the three DNA sequencing options?
A
Whole-genome sequencing, Plasmidsaurus (Plasmids only), Sanger sequencing (only portion of plasmid we care about)

How does Oxford Nanopore Sequencing Technology Work?
A
- DNA strands are pulled through a nanopore
- Miniscule changes in electric current are converted into base reads for each nucleotide
- Produces longer reads then competing technologies
