Explain how we used controls in lab.

Only used positive controls. We didnt have viability controls. The small plasmid was the positive selection control. pJV4 was the positive competency control.

How do you know how many cells received the plasmid?

every “colony” on your plate theoretically comes from a single cell, so by counting colonies we are counting how many cells received the plasmid and were able to grow

Strain generation and screening, scale up, production, downstream processing

inoculating single colonies into liquid culture that can then be screened later
