Transformation procedure

A
  • choose a gene of interest and place it in a plasmid
  • the plasmid can be transformed into E. coli MM294 cells
    • E. coli miseducating into a CaCl2 solution
    • heat shocked–>42’C for 90 seconds
    • the cells are plated on plates containing ampicillin
    • E. coli cells without plasmid= no growth
    • transformed E.coli with plasmid= growth
  • transformed strain can now be isolated/cultured so the plasmid can replicate
  • retrieve the plasmids from the cells and subsequently several copies of gene of interest
