Transformation procedure
A
- choose a gene of interest and place it in a plasmid
- the plasmid can be transformed into E. coli MM294 cells
- E. coli miseducating into a CaCl2 solution
- heat shocked–>42’C for 90 seconds
- the cells are plated on plates containing ampicillin
- E. coli cells without plasmid= no growth
- transformed E.coli with plasmid= growth
- transformed strain can now be isolated/cultured so the plasmid can replicate
- retrieve the plasmids from the cells and subsequently several copies of gene of interest
