Catalase/ Oxidase Test Flashcards
What is the purpose of the catalase test?
to detect if your organism has the enzyme catalase
What are the applications for the catalase test? and examples
to differentiate Gram (+) bacteria Catalase (+) -Staphylococcus - Micrococcus Catalase (-) -Streptococci -Enterococcus
What media is used for the catalase test?
TSA plate
What is the inoculation and incubation guide for the catalase test?
loopful streak, 37C for 48 hours
What is the theory behind the catalase test?
Catalase turns H2O2 into H2O+ O2 (bubbles)
What do you do after incubation for the catalase test?
use a toothpick to put the bacteria on a slide and add one drop of hydrogen peroxide
What are the results of the catalase test?
(+)- Bubbles (-)- no bubbles
What is the purpose of the oxidase test?
to determine if an organism has the enzyme Cytochrome C oxidase
What media is used for the oxidase test?
TSA plate
What is the inoculation and incubation guide for the oxidase test?
loopful streak, 37C for 48 hours
What does one do after incubation for the oxidase test?
use a toothpick put bacteria on a piece of filter paper and then add one drop of oxidase reagent
What are the results of the oxidase test?
(+)- bacteria will be purple of pink (-)- bacteria will remain the same color
Is this catalse positve or negative?
Negative
Is this catalase postive or negative?
Positive
Which organism is postive for Cytochrome C oxidase? How do you know?
PA; the bacteria turned pink
