Tissue Processing Flashcards
1
Q
at what temperature are mohs sections snap frozen
A
-20 to -70 degrees
2
Q
how thick are mohs sections cut
A
.006mm or 6 microns
3
Q
what is the bevel amount in a mohs section
A
45 degrees
4
Q
What should the cryostat temp be at?
A
-20 to-25 degrees
5
Q
Which comes first in the stainer Hematoxylin or Eosin
A
Hematoxylin
6
Q
what are the steps in staining a mohs section
A
- hydration and dehydration (alternate alcohol and water rinses)
2.hematoxylin stain (binds to nucleus)
3.water rinse
4.blueing (put in weakly basic soln and hematoxylin turns blue - differentiation (removing unbound Hematoxylin)
- the blueing and differentiation may repeat
7 Eosin
8 dehydration with alcohol to remove all water
9 clearing with xylene
7
Q
what is the ideal cutting temp of a cryostat
A
-25 degrees centigrade with low room temp low humidity
8
Q
What is the ideal cutting temp for fat
A
-30 centigrade
9
Q
what can high humidity lead to in the cryostat
A
tissue curling and ice crystals forming leading to cracks and fragments in cuts
