How does Grapefruit juice interacts with Simvastatin?
GFJ inhibits cytochrome P450 CYP3A4 enzyme and prevents the metabolism of Simvastatin in the liver which causes an overdose of simvastatin which can lead to side effect such as rhabdomyolysis
Explain why Pitavastatin is not inhibited by GFJ
Pitavastatin is metabolised by CYP2C9 and so unaffected by GFJ interaction
