  2. Circle the spill with disinfectant (microban)
  3. Place dry dry paper towels on the spill
  4. Drench paper towels with disinfectant (microban)
  5. Using gloves, put into a biohazard bag
  6. Repeat procedure
  7. Wash your hands

Flame sterilizing an inoculating needle or loop

A
  • sterilize before use and after using*
  • hold in dominant hand, move end of the wire closed to handle down to the tip until it turns red. Wait 30 seconds before using so it can cool

  • using non dominant hand pick up test tube and using pinky and ring finger take the cap off w/ dominant hand. At a 45 degree angle pass top of tube through the flame then put cap back on

Transferring bacteria from one liquid culture to another

  • hold loop/needle in dominant hand and sterilize it
  • still holding loop, pick up test tube in the non dominant hand and take cap off
  • flame the opening of the test tube and insert the loop into the liquid culture making sure it’s just below the surface
  • reflame the tube again and place the cap on
  • pick up next tube and flame the lip
  • insert the loop just below the surface (at 45 angle)
  • reflame tube and put lid on and put into rack
  • reflame loop/needle

Transferring bacteria to solid media

A
  • flame /sterilize loop
  • pick up test tube in non dominant hand while still holding sterilizing loop and take cap off
  • @45 angle run lip of tube through the flame and insert the loop just below the surface
  • reflame tube and cap it putting it back into tube rack
  • lightly lift petri lid and move loop gently on the surface of the agar
  • close peri dish and reflame sterilizing loop
