Protein Synthesis Inhibitors Flashcards
Aminoglycosides
binds to 30s
interferes w/ initiation complex / adds wrong AA / blocks proofreading
Aminoglycosides examples
gentamicin/tobramicin/amikacin (gta)
Aminoglycosides effective
aerobic g-ve
Aminoglycosides ineffective
g+ve / anaerobic g-ve
Aminoglycosides ADVERSE EFFECTS
nephrotoxicity , ototoxicity , 8th nerve damage , renal failure , tubular necrosis , exacerbates MG
Tetracyclines
bind to 30s / prevents tRNA binding to A site
Tetracyclines examples
doxycycline , minocycline , tigecycline.
Tetracyclines effective
G+ve B. / G-ve r (cholerae) , anaerobes , mycoplasma , spirochetes, chlamydia , rickettsia , protozoa
Tetracyclines ADVERSE
fungal superinfection , PC , damage of teeth & bones , ataxia , phototoxicity , worsens renal failure (not doxy)
Macrolides
binds to 50s / inhibits translocation / pregnancy & child safe
Macrolides examples
erythromycin , clarithromycin , azithromycin
Macrolides effective
G+ve b & c , G-ve r. & c , mycoplasma , spirochetes , chlamydia
Macrolides ADVERSE
NVD , Cramps , cholestatic jaundice (e)
Chloramphenicol
Binds to 50s / inhibits peptidyl transferase + mammalian mitochondrial protein synthesis
Chloramphenicol effective
MRSA , G+ve , G-ve , anaerobes , NM , HI , S. pneumoniae
