1-3 step of prepping a bacterial smear

  1. CLEAN SLIDES remove grase or oil from fingers by washing wiht soap and water or scouring powders such as bon ami.

follwo by water rinse and rinse of 95% alcohol

  1. LABEL SLIDES initials of organism written on either end of the slide with glassware pencil
  2. PREP OF SMEAR avoid thick dense smears, less culture. no light can get through to visualize (from broth or solid)

making a smear from broth medium (after cleaning and labeling slides)

A
  1. sterilize inoculating loop
  2. remove cap with pinky
  3. hold tube on an angle and pass through flame 2-3 times
  4. dip loop
  5. reflame and cap
  6. circular movement spread onto area size of quarter or dime
  7. air dry completely (warming tray)
    8.HEAT FIX pass over flame quickly 2-3 times

making a smear from solid medium (after cleaning and labeling slides)

  1. add a few loop fulls of water to slide
  2. steralize inoculatign loop in thirds (starting at handle end)
  3. pin point amoutn fo sample from slant culture to drop of water spread to nickle size
  4. air dry
  5. heat fix
