- blood film examination : for morphology
- Molecular biology: look for gene JAK2 V617F
- arterial blood gas (pO2) ; bc hypoxia => erythrocytosis
- Epo assay: stimulant
- RBC & plasma vol
- renal function: bc where Epo is released
