What are routes of absorption?

- Enteric coated = intestinal absorption
- Extended release = slower absorption
- Sublingual = buccal avoid 1’st metabolism

A
- What the body does to the the drug.

A
- Passive/facilitated diffusion, active transport, endocytosis

What factors affect absorption?
A
- pH, vascularity, surface area, with food (slower emptying)

What is bioavailability?
A
- Rate and extent to which the drug reaches systemic circulation.
- IV is 100%
- Factors affecting this include first pass metabolism (liver - drug inactive), solubility, chemical instability.

What is distribution?
A
- Drug enters the blood stream and then passes into the extracellular fluid and tissues.
- Factors affecting including blood flow, capillary permeability, plasma/tissue protein binding and liphilicty.

A
- Process of elimination
- First order = rate of metabolic is proportional to drug conc
- Zero order is where the enzyme statured by higher dose
