Neutralization and Inactivation Flashcards
1
Q
What is the purpose of performing a neutralization (inhibition) test?
A
To detect alloantibodies masked by:
- Lewis substances in saliva
- P1 substance - in hydatid cyst fluid and pigeon egg whites
- Sda substance - abundant in urine
- Chido and Rodgers substances - epitopes of C4 (complement)
2
Q
What type of SULFHYDRYL reagents destroys or weakens Kell system antigens?
A
AET and DTT
3
Q
What do the sulfhydryl reagents AET and DTT do?
A
destroys/weakens Kell system antigens
4
Q
What does ZZAP do?
A
Enzyme + DTT destroys Kell antigens and those antigens destroyed by enzymes (Duffy, MNSs)
Can also remove immunoglobulins and complement from RBCs to enhance adsorption
5
Q
What are DTT and 2-ME and what do they do?
A
Sulfhydryl reagents
Destroys/reduces activity of IgM antibodies by cleaving disulfide bonds
