What is an elution?
Process of removing antibodies off of antigens on red blood cells and release it into solution (eluate).
After removal of the AB, test the eluate to determine if the AB is cause of positive DAT in hemolytic transfusion reactions, HDFN, autoimmune and drug related hemolytic anemia
Can complement be eluted off of RBCs?
NO
What kind of solvent is used to elute suspected auto and allo-warm reacting antibodies off of RBCs?
Acid or organic solvents
What is a Lui Freeze/Thaw?
Lui freeze/thaw elution is a technique to remove antibodies from red blood cells when the suspected antibody is anti-A, anti-B, or anti-A,B.
What elution method would you use if they antibody suspected is apart of the ABO system?
Lui freeze thaw
What is the last wash control in elution?
Prior to elution, red blood cells that are coated with antibody are thoroughly washed to remove any residual serum containing other antibodies
The last wash supernatant is kept before performing the elution. It should show NO reactivity with reagent cells.
It ensures the recovered antibody present in the eluate has been released from a bound state on the original cells and not residual unbound antibody remaining as a result of inadequate wash procedure.
If the washes were performed correctly, what would be the result of the last wash control?
Negative
If the last wash control is positive, what would the result be?
Invalid
What is the purpose of an adsorption?
Used to separate multiple antibodies
Remove autoantibody that is masking an alloantibody
Confirm antigen existence on RBC
Confirm antibody specificity
What is an autoadsorption?
An adsorption using patient’s own serum and cells
What is an alloadsorption?
Adsorption using patients serum and other cells
