Absorption
unchanged drug moves from site of administration into systemic circulation
distribution
process of reversible transfer of a drug b/w one location and another
Metabolism
chemical modification of a drug, carried out by enzymes
Excretion
irreversible loss of the drug from the body (kidney)
elimination
loss of drug from plasma via liver
therapeutic range
the range of drug concentrations that has probability of desired effect + low probability of toxic effect
what are the 2 concentrations that the therapeutic range sits in between
- minimum effective concentration
2. minimum toxic concentration
drug half life
the time taken for drug concentration to be reduced from 50% of it’s max concentration
what does the half life of a drug determine
- duration of action of single dosage
- how often a drug needs to be administered to stay within therapeutic range

receptors - agonist
initiates/ enhances normal response

enzymes - competitive inhibitors
drug binds to active site
- substrate can’t bind
- slows/ inhibits enzyme activity
enzymes - non competitive inhibitors
drug binds to enzyme, changing the shape
- prevents binding
- enzyme destroyed
ion channels
drugs bind or block channels to alter functions of proteins
carrier proteins
drugs bind + prevent carrier proteins from moving molecules into the cell
Drug Hypersensitivity
- exaggerated response to a drug perceived as foreign
eg allergic reaction
Adverse Drug Reactions
an unintended/ undesirable effect
- includes hypersensitivity
Drug Interactions (DDI)
occurs when the pharmacological effect of one drug is altered by another
Drug Contraindications
are the conditions that determine when a drug should not be prescribed eg pregnancy
