Microscopy Flashcards
How do light microscopes work?
They work by passing light through the specimen, you can use them to study living cells
How do Electron microscopes work?
They use electrons and pass them through the specimen they have a higher magnification and resolution
What is meant by resolution
It means that the image can be seen more clearly
How to view a specimen using a light microscope.
Take a thin slice of the specimen
Take a clean slide and use a pipette to put one drop of water on it, use the tweezers to put the specimen on it
Add a drop of stain if the specimen is clear place a cover slip with a mounted needle so no air bubbles are made
Use the lowest powered objective lens
Use the coarse adjustment knob so the slide is just underneath it
Put the stage down so that the specimen is in focus
Adjust the focus with the fine adjustment knob go get a clear image
Put a ruler on the stage and use it to measure the diameter of the circular visible area.
What do you do if you need a specimen with a greater magnification?
Swap to a high powered objective lens refocus and recalculate you field of view accordingly
Equation for microscopy
Magnification = image size / real size
Real size = image size / magnification
Image size = magnification * real size
