M4: Types of Agar Plates Flashcards
LB Agar
- aka Lysogeny Broth
- multipurpose, can grow lots of things
- non-selective
- non-differential
- commonly used to grow E. coli
TSA / TSAYE Agar
- aka Trypticase Soy Agar
- non-selective
- non-differential
- base for formulating enriched medias
Blood Agar
- aka BAP
- derivative of TSA w/added sheep blood (at 5-10% concentration)
- non-selective
- differential –> detects hemolytic activity
- enriched for promoting growth of Streptococcus strains
alpha hemolysis
- partial lysis (breakdown) of red blood cells
- shows up as greenish-brown colonies on BAP
beta hemolysis
- complete lysis of red blood cells
- shows up as a clearing around colonies on BAP
gamma hemolysis
- lack of hemolysis
- shows up as white/tan colonies growing on red plate
LB Agar color
pale yellow
TSAYE color
pale yellow
Columbia CNA Agar
- selective –> contains antimicrobial agents that supress gram negative bacteria
- enriched with blood
- differential –> detects hemolytic activity
antimicrobial agents in Columbia CNA Agar (2)
colistin & nalidixic acid)
Columbia CNA Agar color
red
Blood Agar color
red
Chocolate Agar
- aka CHOC or CBA
- non-selective
- non-differential
- enriched with blood cells that have been lysed with heat –> appear brown, promote growth of fastidious pathogenic bacteria
Chocolate Agar color
brown
MacConkey Agar
- selective –> contains crystal violet + bile salts that restrict gram-positive growth
- differential –> lactose + neutral red pH indicator used to differentiate
- often used for intestinal pathogenic microbes (such as Salmonella)
Sorbitol-MacConkey Agar
- aka SMAC
- formulated specifically to detect presence of pathogenic E coli (E coli 0157), which cannot ferment sorbitol
Eosin Methylene Blue Agar
- aka EMB
- selective –> contains methylene bue and eosin that restrict growth of gram pos bacteria
- differential –> lactose fermentation used to differentiate
Mannitol Salt Agar
- aka MSA
- selective –> selects FOR gram pos bacteria via high concentration of sodium chloride
- differential –> can differentiate between strains of staphylococci via fermentation of mannitol + pH indicator
- pathogenic strains ferment mannitol –> lower pH
MacConkey Agar color
pale red/pink
SMAC Agar color
pink
EMB Agar color
dark red
MSA color
red
