M4: Types of Agar Plates Flashcards
1
Q
LB Agar
A
- aka Lysogeny Broth
- multipurpose, can grow lots of things
- non-selective
- non-differential
- commonly used to grow E. coli
2
Q
TSA / TSAYE Agar
A
- aka Trypticase Soy Agar
- non-selective
- non-differential
- base for formulating enriched medias
3
Q
Blood Agar
A
- aka BAP
- derivative of TSA w/added sheep blood (at 5-10% concentration)
- non-selective
- differential –> detects hemolytic activity
- enriched for promoting growth of Streptococcus strains
4
Q
alpha hemolysis
A
- partial lysis (breakdown) of red blood cells
- shows up as greenish-brown colonies on BAP
5
Q
beta hemolysis
A
- complete lysis of red blood cells
- shows up as a clearing around colonies on BAP
6
Q
gamma hemolysis
A
- lack of hemolysis
- shows up as white/tan colonies growing on red plate
7
Q
LB Agar color
A
pale yellow
8
Q
TSAYE color
A
pale yellow
9
Q
Columbia CNA Agar
A
- selective –> contains antimicrobial agents that supress gram negative bacteria
- enriched with blood
- differential –> detects hemolytic activity
10
Q
antimicrobial agents in Columbia CNA Agar (2)
A
colistin & nalidixic acid)
11
Q
Columbia CNA Agar color
A
red
12
Q
Blood Agar color
A
red
13
Q
Chocolate Agar
A
- aka CHOC or CBA
- non-selective
- non-differential
- enriched with blood cells that have been lysed with heat –> appear brown, promote growth of fastidious pathogenic bacteria
14
Q
Chocolate Agar color
A
brown
15
Q
MacConkey Agar
A
- selective –> contains crystal violet + bile salts that restrict gram-positive growth
- differential –> lactose + neutral red pH indicator used to differentiate
- often used for intestinal pathogenic microbes (such as Salmonella)
