What is the meaning of ALT?
Alanine Transaminase (Alanine Aminotransferase)
What is the meaning of SGPT?
Serum Glutamic Pyruvic Transaminase
What is ALT?
It is a cytosolic enzyme that is found in high concentrations in the liver
What is the characteristic of ALT?
It is a liver-specific enzyme
What is hepatocellular injury?
It is the trigger for the release of these enzymes into the circulation
What are the diagnostic significance of ALT?
1) For the evaluation of hepatic disorder
2) Intrahepatic > extrahepatic (source)
3) It has a longer half-life compared to AST
4) ALT can be used to screen blood donors
5) ALT is a more sp and sensitive screening test for post-transfusion hepatitis
What is the method present for ALT?
Enzymatic method
What is the principle of rxn of enzymatic method?
L- Alanine + alpha(sign)-ketoglutarate (via the action of ALT) -> Pyruvate + L- glutamate
Pyruvate + NADH + H^+ (via the action of LDH) -> L- lactate + NAD^+
What are the sources of error for ALT?
1) It is relatively unaffected by hemolysis
2) Stability: 3 - 4 days at ref temp
What are the tubes present for the determination of ALT lvls?
1) ALT + Blank
2) ALT + Standard
3) ALT + Unknown
What is the process (or steps) for ALT?
1) Label test tubes (blank, standard, unknown)
2) Transfer 1,000 uL of ALT working rgnt to each tube and place in a 37 DC H2O bath
3) Add 100 uL sx
4) Read and record absorbance at 5 mins using 340 nm
What is the normal reference range of ALT?
3 - 35 IU/L
