Lipoprotein Lipase Flashcards
Function
hydrolyzes TAGs in chylomicrons and VLDL
cleaves the sn1 and sn3 ester bonds
phospholipase activity to open phospholipase shell
Location
adipose tissue striated muscle lactating mammary gland heart -bound to heparin sulfate carbohydrate chains of glyxocalix at luminal surface
Increased LPL activity
by insulin in adipose tissue
starvation in skeletal muscle and heart
prolactin in lactating mammary gland
Hepatic lipase
instead of LDL in liver lipid hydrolase acts on IDL and HDL2 cleaves both TAGs and phospholipids plays a role in conversion of HDL2 back to HDL3
LDL, HL, pancreatic lipase PL and endothelial lipase
Ser-His-Asp triad in catalytic site
LPL requires C2 apoprotein as cofactor
PL reguires colipase as cofactor
Chylomicronemia syndrome
accumulation of chylomicrons in blood- high TAG conc.
eruptive xanthomas
lipemia retinalis
abdominal symptoms
LPL deficiency
rare, autosomal recessive disorder
clearance of chylomicrons from blood diminished
elevated fasting plasma TAG level
risk of pancreatitis
