LAMP Principles and Mechanism

A

1). Isothermal Amplification: Unlike traditional PCR, which requires thermal cycling (repeated heating and cooling), LAMP is performed at a constant temperature (usually around 60-65°C). This simplifies the equipment needed and speeds up the process.

2). Loop Primers: LAMP uses a set of specially designed primers (typically four to six) to initiate the DNA synthesis. These primers target six distinct regions on the DNA, which allows for high specificity and efficiency.

3). DNA Polymerase: The enzyme used in LAMP is a strand-displacing DNA polymerase, which can synthesize new strands of DNA while simultaneously displacing the old strands. This is essential for the continuous amplification process.
