- Organophosphates
Edrophonium
- Short acting
- Used for the diagnosis of myasthenia gravis
Physostigmine
- Tertiary amine (enters CNS)
- Used to treat glaucoma and as an antidote in atropine overdose
Neostigmine and pyridostigmine
- Quaternary amines (no CNS entry)
- Used to treat ileus, urinary retention, myasthenia gravis, and reversal of non-depolarizing NM blockers (-curiums/-curoniums)
Donepezil and rivastigmine
- Lipid soluble (CNS entry)
- Used to treat Alzheimer disease
What is the equation for volume of distribution?
Dose/Concentration at time 0
What is the equation for half life?
.7 x (Volume of distribution/clearance)
What is the equation for infusion rate?
Clearance x steady state concentration
What is the equation for loading dose?
(Volume of distribution x plasma concentration)/(bioavailability)
What is the equation for maintenance dose?
(Clearance x steady state concentration x dosing interval)/(bioavailability)
