omeprazole, lansoprazole, pantoprazole
PROTON PUMP INHIBITORS - mechanisms of action
irreversible inhibition by covalent modification of the gastric gland parietal cell H+/K+
proton pump = final common pathway
prodrugs - activated in the acidic environment of the canaliculus contributing to their effectiveness and selectivity of action
take PO are absorbed and subsequently delivered to the stomach for activation
PROTON PUMP INHIBITORS - major indictions
peptide ulcers, DU or GU
dyspepsia and GORD
helicobacter pylori infection
amoxicillin, metronidazole, clathithromycin in conjunction with omeprazole
PROTON PUMP INHIBITORS - uncommon indications
zolinger-ellison syndrome
PROTON PUMP INHIBITORS - adverse effects
diarrhoea headache abdominal pain nausea fatigue dizziness
increased risk of C.Diff
result in rebound acid hyper secretion upon discontinuation
