Gene Expression and Function Flashcards
What are the two new techniques being used a ton?
CRISPR and RNAi
What do you want to modify in CRISPR?
Double-strand break
What cleaves in CRISPR system?
Cas-9 protein
How can you repair double strand break?
HR, NHEJ
Why is break/repair useful in CRISPR?
When the gene is repaired, it’s replaced by a different sequence!
What does RNAi stand for? How does it work?
RNA interference. Put it in food and have organisms eat it
Why is RNA useful?
You can have a ton of plates at once with different dsRNA, and see what the function is for everything at once
Why are reporter genes useful?
They code for a protein with a visible protein, so we can use them to tell phenotypically if genes were expressed
What is RT-PCR?
Real-time PCR
What does RT-PCR do? Why is that useful?
Gives fluorescent detection of PCR product in real time.
You can watch a PCR reaction as it unfolds.
What does RT-PCR do? Why is that useful?
Gives fluorescent detection of PCR product in real time.
You can watch a PCR reaction as it unfolds.
Why is it good to be able to watch PCR as it unfolds?
If you have two mRNA sequences, you can see which one amplifies faster
What do microarrays do?
Scan for fluorescent signals to make images.
