a mutation results in a different sequence of amino acids, therefore different R groups, resulting in changes to ionic hydrogen and disulphide bonds, changes in shape of active site causing a change in enzyme activity

enzymes lower the activation energy of reactions in the clotting pathway therefore speeding it up, enzymes are not destroyed by the reaction they catalyse and speed it up
