What is the mechanism of action of ondansetron?
5-HT3 antagonist (acts on CTZ)
What is the mechanism of action of promethazine?
Histamine H1 receptor antagonist (does not affect CTZ)
What is the mechanism of action of scopolamine?
Non-selective muscarinic antagonist (acts in vestibular nuclei via M1 receptors)
What is the mechanism of action of aprepitant?
Neurokinin-1 receptor antagonist (acts on CTZ)
What is the mechanism of action of metoclopromide?
Dopamine D2 receptor antagonist (acts on CTZ)
What are the drugs used to treat acute migrane attacks?
- Sumatriptan
- Naratriptan
- Ergotamine
What is the mechansim of action of sumatriptan?
5-HT1B/D/F agonist:
- 5-HT1B = Vasoconstrction
- 5-HT1D/F = Nociceptor inhibition
What are the differences between sumatriptan and naratriptan?
Naratriptan:
- Crosses BBB
- Longer duration of action
- Fewer cardiac side-effects
What are the drugs used to prevent migrane attacks (prophylaxis)?
- Propanolol (non-specific β-adrenoreceptor antagonist)
- Topiramate
