drugs included in CYTOPROTECTIVE/MUCOSAL protective drugs:
- sucralfate
- misoprostol
- colloidal bismuth
mechanism of action of sucralfate:
- aluminum sucrose sulfate
2. bind to injured tissues → forms a protective coating over ulcer beds
pharmacokinetics of sucralfate:
- acc. healing & reduce recurrence
2. administration = MUST be taken → 4x daily
indications of sucralfate:
- duodenal ulcers
2. prevention of stress ulcers
side effects of sucralfate:
- low toxicity
2. drug-interactions (need acidic pH, cannot be combined with PPI, H2- blockers or antacids)
mechanism of action of misoprostol:
- analog of PGE1
2. increase mucosal protection & inhibit acid secretion
indications of misoprostol:
reducing risk of ulcers in users of NSAIDs
side effects of misoprostol:
- gastrointestinal upset
- diarrhea
(this is a poorly tolerated drug!)
CONTRAindication of misoprostol:
PREGNANCY
mechanism of action of colloidal bismuth:
- forms protective coating on ulcerated tissue
- stimulate mucosal protective mechanism
- antimicrobial effect
- sequestration of enterotoxins
pharmacokinetics of colloidal bismuth:
used in quadruple therapy to heal peptic ulcers
indications of colloidal bismuth:
- peptic ulcers
2. reduce stool frequency & liquidity in infectious diarrhea
side effects of colloidal bismuth:
black stools
