BMSC 240 Lecture 9-10 Flashcards
Serial Dilution
divide by the volume plated, multiply by the volume of the tube ,divide by the volume placed in that tube from the previous.
Antibiotic
kills bacteria
bacteriolytic-
cause decrease in OD
Bacteriostatic
causes growth curve to level off
toxic dose
amount harmful to humans
Antimicrobial targets
inhibition of cell wall synthesis, inhibition of protein synthesis, inhibition of nucleic acid synthesis, injury to plasma membrane.
Cell wall inhibitors
i.e penecillins and cephalaphorins(lytic). All contain B-lactan ring. irreversible bind and and inhibit transpeptidase in the periplastmic space.
Protein synthesis inhibitor
i.e tetracycline. affects ribosomes/ mRNA and tRNA.
Plasma membrane Damage
i.e. polysporin. polymyxins and daptomycin.
Nucleic Acid synthesis inhibition
Rifamycin-inhibit mRNA synthesis. counteract lyprosy,
Quinolones-inhibit DNA gyrase and DNA replication… cipofloxin.
Metabolic inhibitors
blocks an enzyme needed to produce folate.
ways to misuse antibiotics
- ) discontinuing them before you the pathogen is eradicated.
- ) using poorly chosen antibiotic
- ) use of antibiotic against viral infection
- )
What are the 5 modes of resistance
- ) drug modification/destruction.
- ) pathway protection
- ) Target alteration
- ) Rapid efflux
- ) Alternative pathways
What is a plasmid?
small, extrachromosomal ring of DNA that can be transferred to other individuals or species.
