BMSC 240 lecture 8 Flashcards
What physical factor affect growth?
Temperature, salinity, nutrients, pH
Where do facultative halophiles live in your body?
up your nose
Complex media
Wearenotawareofevery chemicalinthemedia
– Partiallydigestedbeef
– Groundupdriedyeast
Defined Media
Everychemicalinthemediais
knownandquantified
– Sugars,aminoacidsandinorganic nutrients
– Canbetailoredtorequirementsof onespecies
Media broth
-grow large quantities of culture, difficult to check for purity
media plate
readily score and separate colonies. doesn’t store well
slants
miniplates. for storage of smaller volumes. difficult to check for purity
semi solid deeps
can score motility and oxygen requirements, also storage. difficult to check to check for purity
What is a nutrient agar plate suitable for?
Suitable for non-fatsitious organisms
Selective media
will encourage the growth of some bacteria while preventing the growth of others
differential media
will produce different visual cues as to the type of organism growing.
Enrichment media
is a type of selective media used to allow proliferation of rare bacteria in mixed population (soil, feces)
MacConkey Agar
Selective and differential. Selects against gram positive using bile salts and crystal violet. Fermentors turn Purple.
Mannitol Salt Agar
selective and differential. fermentors turn yellow and non fermentors turn red.
Blood Agar plates
Test for hemolytic activity.Alpha is discolouration in or under the colony. Beta-is clearing. Gammma-no hemolysis.
