Bacterial Culture Methods Video Flashcards
What are the 4 criteria for cell growth?
nutritional requirements, time, temperature, atmospheric conditions
What do you use to grow bacteria?
agar because has all the nutritional needs for the bacteria to grow
How long do you grow bacteria?
usually up to 4 days
What temperature is good for most bacterial growth?
body temperature so 35-37F
42 F for ???
yeasts grow at 30 F
What types of bacteria grow in a CO2 incubator?
A. aerobic
B. microaerophilic
C. anaerobic
A. aerobic
B. microareophilic
Where do we grow anaerobic bacteria?
in sealed jars/containers to seal off oxygen
What is the purpose of gas pack in anaerobic bacterial growth?
produces Hydrogen gas to form with the oxygen that may get in sealed jar to form water
What is the purpose of gram staining?
this will show us our bacterial shape so we can use a more specific antibiotic to kill it
What are the 4 steps of gram staining?
- crystal violet stain
- apply iodine
- apply the acetone alcohol
- apply safranin
What is the purpose of the crystal violet stain?
to stain everything on the slide
What is the purpose of applying iodine?
to act like a binding agent for crystal violet to all and only gram + organisms
What is the purpose of applying acetone alcohol?
decolorize the slides and take off excess crystal violet that is present
What is the purpose of applying safranin?
to stain gram - organisms
