MOA: Links and inhibits haemoglobin, therefore blocking detoxification

MOR: Mutation of gene, Pfcrt, which encodes a transmembrane protein in the parasite’s food vacuole

AE:

  • Retinal degredation
  • Skin itching in West Africans
  • Hypotension, heart block , cardiac arrest
  • Suicide attempts

-Drug of choice for area of the world w/ no chloroquine resistance

Mefloquine

-AE

A

-Active against most strains of chloroquine - resistant P. falciparum

AE:

  • Decreased coordination–> important for pilots
  • worsen psychiatric disorders–> depression

use with caution in patients using beta blocker
